Active Ingredient

Akorn Tolazine Injection contains blocking agent Tolazoline for reversal of the effects of sedation and analgesia caused by alpha-2 adrenergic agonists such as xylazine.
Each mL contains Tolazoline hydrochloride equivalent to 100 mg base activity

Actions

Clinical Pharmacology: Tolazoline belongs to the synthetic group of alpha-adrenergic blocking agents known as the imidazole derivatives. It is a mixed alpha-1 and alpha-2 adrenergic receptor antagonist that competitively inhibits alpha-adrenoceptors. Tolazoline is also a direct peripheral vasodilator that decreases the peripheral resistance and increases venous capacitance.
Xylazine is an alpha-2 adrenergic agonist with sedative and alangesic properties related to central nervous system depression. Administration of Akorn Tolazine Injection reverses xylazine’s central nervous system depressant effects resulting in rapid recover from sedation. The competitive blocking of the alpha-2 adrenergic receptor by Akorn Tolazine Injection displaces xylazine from these sites and thereby reapidly cancels its effect.
Onset of arousal is usually apparent within 5 minutes of Akorn Tolazine Injection administration, depending on the depth and duration of xylazine induced sedation.

Indications

Akorn Tolazine Injection should be used when it is desirable to reverse the effects of sedation and analgesia caused by alpha-2 adrenergic agonists (eg xylazine).

Dosage and Administration

The Akorn Tolazine Injection dose is 2-4 mg/kg body weight (2-4 mL/100 kg) to reverse the sedative effects of xylazine. The calculated dose of Akorn Tolazine Injection should be given slowly by intravenous injection and titrated to effect. Administration rate should be approximately 1 mL/second.
Horses 2 - 4 mg/kg i.v. (2 – 4 mL i.v. per 100 kg)
Cattle 2 - 4 mg/kg i.v. (2 – 4 mL i.v. per 100 kg)
Deer 2 - 4 mg/kg i.v. (2 – 4 mL i.v. per 100 kg)
Sheep and Goats 2 - 4 mg/kg i.v. (0.2 – 0.4 mL i.v. per 10 kg)
Dogs 2 - 4 mg/kg i.v. (0.2 – 0.4 mL i.v. per 10 kg)
Cats 2 - 4 mg/kg i.v. (0.05 – 0.1 mL i.v. per 2.5 kg)
